Administration of a radioisotopic compound by infusion over a period of time greater than two hours, preferably greater than twelve hours, greatly increases the maximum radioactivity that accumulates in the target cell. The efficacy of the administration of the radiolabeled compound can be increased about five times higher than prior bolus injection or short infusion methods. This method enhances the tumor to background ratio by increasing the actual radioligand accumulated inside the tumor cells. This technique works for any radiolabeled compound whose cellular uptake is limited by a cellular process of either binding to a cellular receptor or to a transport protein. Once the radiolabeled compound is bound and internalized, the ability of an unlabeled compound to compete with the radioligand is markedly decreased. The primary factor governing residence time after internalization is the physical half-life of the radioisotope, not biologic half-life.

Le fait d'administrer par perfusion un composé à base de radio-isotope pendant plus de deux heures, plus de douze heures, de préférence, augmente notablement la radioactivité maximale qui s'accumule dans la cellule cible. On peut multiplier par cinq l'efficacité de l'administration de ce composé radiomarqué, comparé à celle des injections pratiquées jusqu'à maintenant, en l'occurrence les injections bolus et les perfusions "courtes". Cette technique permet d'élever le rapport tumeur/ radioactivité naturelle par renforcement du radioligand réel accumulé dans les cellules tumorales. Cette technique est applicable à tous les composés radiomarqués dont l'assimilation cellulaire est limitée par un processus cellulaire donné, à savoir, la fixation à un récepteur cellulaire ou à une protéine de transport. Une fois le composé radiomarqué fixé et internalisé, l'aptitude d'un composé non marqué à concurrencer le radioligand est nettement réduite. Le facteur premier commandant le temps de séjour après internalisation est la période radioactive du radio-isotope, du point de vue de la physique, et non sa demi-vie, biologiquement parlant.
