In a method for the determination of an analyte in a sample involving utilizing biospecific affinity reactions, and comprising the following steps: i) forming a complex comprising: Reactant I--Analyte'-- Reactant*, where a) Reactant* and Reactant I exhibit biospecific affinity to the analyte, b) Reactant* is analytically detectable, c) Analyte' is the analyte or an analyte related reactant, and subsequently ii) determining the detectable signal from Reactant* in the complex (sample value), and iii) obtaining the amount of analyte in the sample by comparing the sample value with one or more calibrator values, each of which corresponds to a standard amount of analyte, either (i) the calibrator or (ii) a binder for the calibrator has been bound before the determination of the calibrator value to a matrix which is insoluble in the liquid medium in which binding of Reactant* to the calibrator occurs, wherein, when a binder for the calibrator has been bound to the matrix, calibrator is added or calibrator pre-deposited in the matrix is released at the determination of calibrator value. The invention also relates to a device and a test kit for performing the method.
On décrit un procédé pour déterminer un analysat dans un échantillon en mettant en oeuvre des réactions d'affinité biospécifique. Le procédé consiste à: i) former un complexe contenant un réactif I, un analysat' et un réactif*, dans lequel a) le réactif* et le réactif I présentent une affinité biospécifique pour l'analysat, b) le réactif* est décelable par analyse, c) l'analysat' est ledit analysat ou un réactif lié à l'analysat; ii) déterminer dans le complexe le signal décelable émanant du réactif* (valeur de l'échantillon); et iii) déterminer la quantité d'analysat dans l'échantillon en comparant la valeur de l'échantillon avec une ou plusieurs valeurs d'étalonnage correspondant chacune à une quantité d'analysat de référence. Un étalonneur ou un liant pour l'étalonneur sont liés avant la détermination de la valeur d'étalonnage relativement à une matrice insoluble dans le milieu liquide dans lequel s'effectue la liaison du réactif* à l'étalonneur. Lorsqu'un liant pour l'étalonneur est lié à la matrice, l'étalonneur est ajouté ou l'étalonneur préalablement placé dans la matrice est libéré au moment de déterminer la valeur de l'étalonneur. L'invention concerne également un dispositif et une trousse d'analyse servant à réaliser le procédé.
