A method for identifying a compound for modulating the immune response in a first type of mammal to a superantigen, comprising the steps of 5 (7) providing a cell expressing a MHC Class II molecule derivable at least in part from the first type, or cells comprising such a cell or cells (8) exposing the cell or cells provided in step (1) to a test compound (9) before, after or during step (2) exposing the cell or cells to the superantigen (10) determining whether the test compound modulates a response of the cell or cells to exposure to the superantigen (11) determining whether the test compound modulates a response of such a cell or cells to exposure to a conventional antigen, and (12) selecting a compound which modulates a response to the superantigen but does not modulate the response to exposure to the conventional antigen. The test compound may be a monoclonal antibody reactive with MHC Class II molecules. A method of treating an individual with or at risk of superantigen exposure/poisoning comprising administering to the individual the antibody TDR3 1.1 or molecule having the binding specificity of TDR31.1.

L'invention concerne un procédé d'identification d'un composé de modulation de la réponse immunitaire chez un premier type de mammifère, vis-à-vis d'un superantigène, procédé comprenant les étapes suivantes : (7) préparation d'une cellule exprimant une molécule MHC classe II, dérivable, au moins partiellement, du premier type, ou des cellules comprenant une ou plusieurs de telles cellules, (8) exposition de la/ou des cellules fournies à l'étape (1) à un composé test, (9) avant, après, ou durant l'étape (2), exposition de la/ ou des cellules au superantigène, (10) détermination permettant de savoir si le composé test module une réponse de la/ou des cellules à l'exposition au superantigène, (11) détermination permettant de savoir si le composé test module une réponse d'une telle cellule, ou de telles cellules, à une exposition à un antigène conventionnel, et (12) sélection d'un composé modulant une réponse au superantigène, mais ne modulant pas la réponse à l'exposition à l'antigène conventionnel. Le composé test peut être un anticorps monoclone réactif avec des molécules MHC classe II. L'invention concerne en outre un procédé de traitement d'un sujet avec/ ou risque d'exposition/empoisonnement par un superantigène, procédé consistant à administrer au sujet l'anticorps TDR31.1 ou une molécule ayant la spécificité de liaison de TDR31.1.
