The invention provides methods and apparatus for carrying out multiple amplification reactions in a single reaction chamber by successive cycles of loading reaction mixture, amplifying, and removing spent reaction mixture in a fluidly closed reaction system. In particular, the present invention allows amplification of a plurality of target polynucleotides from a single sample by carrying out under closed-loop control successive amplifications of different target polynucleotides from different portions of the sample.

L'invention concerne des procédés et un dispositif servant à exécuter de multiples réactions d'amplification dans une seule chambre de réaction par une succession de cycles de chargement du mélange réactionnel, d'amplification et d'évacuation du mélange réactionnel épuisé dans un système réactionnel fermé eu égard à la circulation des fluides. L'invention permet notamment l'amplification d'une pluralité de polynucléotides cibles à partir d'un seul échantillon grâce à l'exécution par commande en boucle fermée d'amplifications successives de différents polynucléotides cibles à partir de différentes fractions de l'échantillon.
