The present invention relates to soluble guanylate cyclase (sGC) enzymes. In particular it provides a cell-based assay system which allows to detect with high efficiency and sensitivity the soluble guanylate cyclase enzymatic activity. In a preferred embodiment, the invention provides a method of screening molecules that interact with sGC by stimulating or inhibiting its enzymatic activity and that can be used in the diagnosis or therapy of sGC- mediated dysfunctions or diseases. The invention further provides gene- constructs, vectors, and cells for use in said method.

La présente invention concerne des enzymes guanylates cyclases solubles (sGC). Plus particulièrement, cette invention concerne un système de dosage biologique fondé sur des cellules permettant de détecter, avec une grande efficacité et une grande sensibilité, l'activité enzymatique des guanylates cyclases solubles. Dans un mode de réalisation privilégié, cette invention concerne un procédé permettant de cribler des molécules interagissant avec les sGC par stimulation ou inhibition de leur activité enzymatique et pouvant être utilisées pour diagnostiquer ou traiter des maladies ou des dysfonctions associées aux sGC. Cette invention concerne également des gènes chimères, des vecteurs, et des cellules pouvant être utilisés dans le procédé susmentionné.
