This invention relates to methods for treatment, diagnosis and prevention of bone disease and comprises methods including measurement and modulation of sympathetic tone and leptin activity. Alteration of sympathetic tone in bone disease can be accomplished by decreasing or increasing leptin synthesis, leptin receptor synthesis, leptin binding to the leptin receptor, and leptin receptor activity. Alteration of sympathetic tone in bone disease can also be accomplished in the foregoing manner, in combination with traditional sympathetic nervous system agonists and/or antagonists, such as, but not limited to, a dopamine .beta. hydroxylase antagonist or a .beta. adrenergic antagonist for treatment or prevention of osteoporosis.
L'invention concerne des procédés de traitement, de diagnostic et de prévention de maladie osseuse comprenant des procédés qui consistent en une mesure et une modulation du tonus sympathique et de l'activité de leptine. Lors de maladie osseuse, il est possible de réaliser un changement du tonus sympathique par diminution ou augmentation de la synthèse de leptine, de la synthèse de récepteur de leptine, de la liaison de leptine à son récepteur, et de l'activité de récepteur de leptine. Ce changement peut être aussi réalisé de la manière précédente en combinaison avec des agonistes et/ou des antagonistes classiques du système nerveux sympathique, notamment, sans y être limité, avec un antagoniste de dopamine .beta. hydroxylase ou un antagoniste .beta. adrénergique, pour le traitement ou la prévention de l'ostéoporose.
