The invention provides a method of inducing a mucosal immune response in a subject, comprising administering to the mucosa of the subject an amount of antigen-encoding DNA effective to induce a mucosal immune response complexed to a transfection-facilitating lipospermine or a lipospermidine. In the method of inducing a mucosal immune response, the antigen-encoding DNA can encode an antigen that is expressed on the surface of infected cells during the course of infection. DNA encoding the envelope glycoproteins of viral pathogens is used in the present method. Lipospermines and lipospermidines are bifunctional molecules consisting of one or more hydrophobic chains covalently linked to a cationic grouping in which there is coordination of three or more amide hydrogens with a phosphate oxygen of the DNA chain forming an ionic charge complex. One preferred example of a lipospermine is DOGS (droctadecylamidoglycylspermine). The invention also provides a composition, comprising an amount of DNA encoding an envelope antigen or envelope- associated antigen of a pathogen complexed to a lipospermine. More specifically, the invention provides a composition, comprising an amount of DNA encoding an envelope antigen of HIV complexed to a lipospermine.
Procédé permettant d'induire une réponse immunitaire des muqueuses chez un sujet, qui consiste à administrer à la muqueuse du patient une quantité d'ADN codant un antigène, efficace pour induire une réponse immunitaire des muqueuses, ledit ADN étant mélangé à une lipospermine ou lipospermidine facilitant la transfection. Dans ledit procédé, l'ADN codant un antigène peut coder un antigène qui est exprimé à la surface de cellules infectées au cours de l'infection. De l'ADN codant les glycoprotéines d'enveloppe de pathogènes viraux est utilisé dans la présente invention. Les lipospermines et lipospermidines sont des molécules bifonctionnelles constituées d'une ou plusieurs chaînes hydrophobes liées de manière covalente à un groupement cationique dans lequel il existe une coordination de trois ou plusieurs hydrogènes d'amide avec un oxygène-phosphate de la chaîne d'ADN formant un complexe à charge ionique. Un exemple préféré de lipospermine est DOSG (droctadécylamidoglycylspermine). Ladite invention concerne également une composition comprenant une quantité d'ADN codant un antigène d'enveloppe ou un antigène associé à une enveloppe d'un pathogène mélangé avec une lipospermine. Plus spécifiquement, la présente invention concerne une composition comprenant une quantité d'ADN codant un antigène d'enveloppe de VIH mélangé à une lipospermine.
