The present invention provides a method of inhibiting angiogenesis within a tissue by providing exogenous PEDF to cells associated with the tissue. The presence of exogenous PEDF inhibits angiogenesis within the tissue, in part by interfering with the ability of vascular endothelia to expand within the tissue. The invention also provides a method for determining the severity of a tumor by assaying for the presence of PEDF within the tumor. The invention further provides a method of inhibiting endothelial cell migration, a method of stimulating the growth of hair in a mammal, a method for inhibiting the growth of a tumor, a method of inducing differentiation of a neuroblastoma cell, a method of slowing the growth of a neuroblastoma cell, and method of treating ischemic retinopathy in a mammal. To facilitate the inventive methods, the present invention provides pharmaceutical compositions including sources of PEDF.
L'invention concerne un procédé permettant d'inhiber l'angiogenèse dans un tissu au moyen d'un apport de PEDF (facteur dérivé de l'épithélium pigmentaire) exogène aux cellules associées à ce tissu. La présence de PEDF exogène inhibe l'angiogenèse dans le tissu, notamment en interférant avec la capacité de développement de l'endothélium vasculaire dans le tissu. L'invention concerne également un procédé d'évaluation du degré de gravité d'une tumeur, consistant à rechercher la présence de PEDF dans la tumeur. L'invention concerne en outre un procédé permettant d'inhiber la migration des cellules endothéliales, un procédé permettant de stimuler la pousse des cheveux/poils chez un mammifère, un procédé permettant d'inhiber la croissance d'une tumeur, un procédé permettant d'induire une différentiation d'une cellule de neuroblastome, un procédé permettant de ralentir le développement d'une cellule de neuroblastome, et un procédé permettant de traiter les rétinopathies ischémiques chez un mammifère. L'invention concerne également des compositions pharmaceutiques comprenant des sources de PEDF, contribuant à la mise en oeuvre de ces procédés.
