The invention concerns in particular human chondrocyte cell compositions, and methods for preparing them and their uses. More particularly, the invention concerns the production of autologous human chondrocyte suspensions, using recombinant enzymes and/or media compatible with pharmaceutical use. The invention further concerns methods and compositions for cryopreservation of chondrocytes, in particular in the absence of DMSO. The resulting chondrocytes can be used in vivo for restoring affected cartilaginous structures, such as post-traumatic cartilaginous defects or osteochondritis dissecans of the knee or, more generally, for treating and repairing clinically significant defects of the cartilage, especially articular cartilage.
La présent invention concerne des compositions de cellules chondrocytaires, notamment humaines, et des méthodes pour leur préparation et leurs utilisations. L'invention décrit plus particulièrement la production de suspensions de chondrocytes humains autologues, utilisant des enzymes recombinantes et/ou des milieux compatibles avec un usage pharmaceutique. L'invention décrit également des méthodes et compositions pour la congélation de chondrocytes, notamment en l'absence de DMSO. Les chondrocytes produits peuvent être utilisés in vivo pour restaurer des structures cartilagineuses affectées, telles que des défauts cartilagineux post-traumatiques ou l'ostéochondrite disséquante du genou ou, plus généralement, pour le traitement et la réfection des défauts cliniquement significatifs du cartilage, notamment du cartilage articulaire.
