The invention concerns methods and constructs for producing retroviral particles, in vitro, ex vivo or in vivo. It also concerns the use of said methods and constructs for transferring nucleic acids into cells. More particularly, the invention concerns a composition comprising the whole set of genetic elements required for constituting a retroviral particle, incorporated in one or several defective recombinant adenoviruses for all or part of the regions E1 and E4 at least (adenoviral/retroviral chimera vectors).

L'invention concerne des méthodes et constructions permettant la production de particules rétrovirales, in vitro, ex vivo ou in vivo. Elle concerne également l'utilisation de ces méthodes et constructions pour le transfert d'acides nucléiques dans les cellules. L'invention concerne plus particulièrement une composition comprenant l'ensemble des éléments génétiques nécessaires à la constitution d'une particule rétrovirale, incorporés dans un ou plusieurs adénovirus recombinants défectifs pour tout ou partie des régions E1 et E4 au moins (vecteurs chimériques adéno-rétro).
