Compositions and methods for enhancing disease resistance to a pathogen in a plant are provided. Methods of the invention comprise stably transforming a plant with an antisense nucleotide sequence for a gene encoding an enzyme in the C-5 porphyrin metabolic pathway and operably linking said antisense sequence to a pathogen-inducible promoter, such that invasion of a cell by a pathogen elicits a hypersensitive-like response that results in confinement of the pathogen to cells of initial contact. Transformed plants and seeds are provided. Nucleotide sequences encoding a wild-type maize urod gene useful in the present invention and the amino acid sequence for the protein encoded thereby are provided. These compositions are also useful for regulating cell death in specifically targeted tissues. A maize lesion mimic, dominant mutant phenotype, designated Les22, and the molecular basis for its manifestation are also provided.

L'invention porte sur des compositions et méthodes renforçant la résistance de végétaux aux maladies dues à un agent pathogène. Lesdites méthodes consistent à transformer de manière stable une plante à l'aide de la séquence nucléotidique antisens d'un gène codant pour une enzyme du mécanisme métabolique C-5 de la porphyrine, et liant fonctionnellement ladite séquence antisens à un promoteur inductible par l'agent pathogène, de manière à ce que l'invasion d'une plante par un agent pathogène suscite une réponse du type hypersensibilité se traduisant par le confinement de l'agent pathogène aux cellules de contact initial. L'invention porte également sur des plantes et semences transformées, sur des séquences nucléotidiques codant pour le gène urod du maïs sauvage utile dans le cadre de ladite invention, et sur la séquence d'acide aminé de la protéine ainsi codée. Lesdites compositions régulent la mort cellulaire dans des tissus spécifiquement ciblés. L'invention porte en outre sur un phénotype mutant dominant, dit Les22 mimétique d'une lésion du maïs et sur les bases moléculaires de sa manifestation.
