The present invention provides methods for attenuating gene expression in a cell, especially in a mammalian cell, using gene-targeted double stranded RNA (dsRNA), such as a hairpin RNA. The dsRNA contains a nucleotide sequence that hybridizes under physiologic conditions of the cell to the nucleotide sequence of at least a portion of the gene to be inhibited (the ~target~ gene).
L'invention concerne des procédés pour atténuer l'expression génique dans une cellule, notamment dans une cellule de mammifère, au moyen d'ARN (ARNds) double brin ciblé sur le gène, tel qu'un ARN en épingle à cheveux. L'ARNds contient une séquence nucléotidique qui hybride, sous certaines conditions physiologiques de la cellule, en une séquence nucléotidique d'au moins une partie du gène à inhiber (le gène "cible").
