Methods and compositions to label oligonucleotides and analogs directly on a solid-support having the structure (I), where S is a solid-support, A is a cleavable linker, X is a moiety with three or more attachment sites, L is a label, Y is a nucleophile, i.e. O, NH, NR or S, and P1 is an acid cleavable protecting group are provided. The labelled solid-support is reacted in a cyclical fashion to synthesize a labelled oligonucleotide on a solid-support in the 5' to 3' direction, having the structure (II). Labelled oligonucleotides are also synthesized by reacting: (i) a label reagent bearing functionality consisting of carboxylic acid, sulfonic acid, phosphonic acid, or phosphoric acid, (ii) an oligonucleotide on solid support with nucleophilic functionality, and (iii) a coupling reagent, whereby an ester, amide, thioester, sulfonamide, sulfonate, phosphonate, phosphoramidate, phosphorothioate, or phosphate bond is formed. The labelling reaction may be conducted at label sites including the 5' terminus, the 3' terminus, a nucleobase, an internucleotide linkage, a sugar, amino, sulfide, hydroxyl, and carboxyl.

L'invention concerne des méthodes et des compositions permettant de marquer des oligonucléotides et des analogues directement sur un support solide de structure (I), dans laquelle S est un support solide, A est un liant clivable, X est une fraction à trois sites de fixation au moins, L est un marqueur, Y est un nucléophile, c'est-à-dire O, NH, NR ou S, et P1 est un groupe de protection acide clivable. On fait réagir le support solide marqué de façon cyclique de manière à synthétiser un oligonucléotide marqué sur un support solide dans les sens 5' et 3', de structure (II). On produit également par synthèse des oligonucléotides marqués en faisant réagir : (i) une fonctionnalité porteur d'un réactif de marquage constituée d'acide carboxylique, d'acide sulfonique, d'acide phosphonique ou d'acide phosphorique, (ii) un oligonucléotide sur support solide à fonctionnalité nucléophile, et (iii) un réactif de couplage, ce qui permet de former un liaison ester, amide, thioester, sulfonamide, sulfonate, phosphonate, phosphoramidate, phosphorothioate, ou phosphate. La réaction de marquage peut être conduite au niveau de sites de marquage comprenant la terminaison 5', la terminaison 3', une nucléobase, une liaison internucléotide, un sucre, un amino, un sulfure, un hydroxyle et un carboxyle.
