The invention provides methods and pharmaceutical compositions thereof for treating injury to the central nervous system (CNS). The method includes administering to the CNS of a patient suffering from such an injury a therapeutically effective amount of an inhibitor of acetylcholinesterase (AChE) production immediately following the injury. The methods use as the inhibitor a synthetic nuclease resistant antisense oligonucleotide or ribozyme that is directed against an accessible domain of the AChE mRNA brain variant. The treatment downregulates AChE production and thereby activity. The injury to the CNS may be a head injury (closed or open), brain injury, or spinal cord trauma or other trauma to the CNS.
L'invention concerne des procédés et des compositions pharmaceutiques connexes permettant de traiter une lésion du système nerveux central (CNS). Le procédé consiste à administrer au système nerveux central d'un patient souffrant d'une telle lésion une quantité thérapeutiquement suffisante d'un inhibiteur de la production d'acétylcholinestérase (AChE) tout de suite après la lésion. Le procédé utilise comme inhibiteur un oligonucléotide ou un ribozyme anti-sens qui résistent à la nucléase synthétique et sont dirigés contre une partie accessible de l'ARN messager de l'AChE de la variante cérébrale. Le traitement régule de manière négative la production d'AChE et par conséquent leur activité. La lésion du système nerveux central peut être une blessure au niveau de la tête (ouverte ou fermée), une lésion cérébrale, ou un traumatisme de la moelle épinière ou d'autres lésions du système nerveux central.
