Methods are provided for locally administering an agent to a host. Specifically, the subject methods provide for the local administration of an agent to an interstitial space of a host. In the subject methods, an agent is retroinfused into a vessel of a host, typically a vein, under conditions sufficient for the agent to enter an interstitial space of the host proximal to the vessel location into which the agent is retroinfused. In practising the subject methods, the agent is administered to the host in combination with the production of vascular stress upstream of the site of administration and adjacent to the target interstitium, where the vascular tissue stress is sufficient to provide for transport of the agent from the vascular site of deposition into the target interstitial space. In a preferred embodiment, the agent is retroinfused at a pressure sufficient to provide for mechanical stress on the vessel proximal to the target interstitial space. Also provided are kits for use in practising the subject methods. The subject invention finds use in the local administration of a variety of different agents for treatment of a variety of different disease or other conditions.
L'invention concerne des méthodes d'administration locale d'un agent à un hôte. Plus précisément, ces méthodes concernent l'administration locale d'un agent dans une espace interstitiel d'un hôte. Ces méthodes consistent à introduire un agent par perfusion rétrograde dans un vaisseau d'un hôte, en général une veine, dans des conditions permettant à l'agent d'entrer dans un espace interstitiel de l'hôte à proximité de l'emplacement du vaisseau dans lequel l'agent est introduit par perfusion rétrograde. Dans la pratique, l'agent est administré à l'hôte avec production d'une contrainte sur le tissu vasculaire, en amont du site d'administration et adjacent à l'interstice cible, cette contrainte sur le tissu vasculaire étant suffisante pour permettre le transport de l'agent depuis le site vasculaire de dépôt jusque dans l'espace interstitiel. Dans un mode de réalisation préféré, l'agent est introduit par perfusion rétrograde à une pression suffisante pour fournir une contrainte mécanique sur le vaisseau, à proximité de l'espace interstitiel cible. L'invention concerne également des trousses destinées à être utilisées pour mettre en oeuvre ces méthodes. Cette invention est utile pour l'administration locale d'une variété d'agents différents destinés au traitement d'une variété de maladies différentes ou d'autres états.
