Alternative and/or improved methods are described for the exact removal of a selected subfragment from a DNA molecule by intrachromosomal recombination between two directly repeated DNA sequences using a rare-cleaving double stranded break inducing DNA endonuclease expressed under control of a micro-spore specific promoter. These methods can be applied for the exact exchange of a target DNA fragment for a DNA fragment of interest in plant cells and plants.

L'invention concerne des procédés alternatifs et/ou améliorés pour l'enlèvement exact d'un sous-fragment sélectionné à partir d'une molécule d'ADN par recombinaison intrachromosomique entre deux séquences d'ADN directement répétées à l'aide d'une ADN endonucléase induisant une cassure double brin à clivage rare, exprimée sous le contrôle d'un promoteur spécifique d'une microspore. Ces procédés peuvent être appliqués pour l'échange exact d'un fragment d'ADN cible contre un fragment d'ADN d'intérêt dans des cellules et des plantes.
