A highly sensitive assay is disclosed which combines immunomagnetic enrichment with multiparameter flow cytometric and immunocytochemical analysis to detect, enumerate and characterize carcinoma cells in the blood. The assay can detect one epithelial cell or less in 1 ml of blood and has a greater sensitively than conventional PCR or immunohistochemistry by 1-2 orders of magnitude. In addition, the assay facilitates the biological characterization and staging of carcinoma cells.
On divulgue l'invention d'un essai très sensible, qui combine un enrichissement immunomagnétique à l'aide de la cytofluorométrie en flux à plusieurs paramètres et l'analyse immunocytochimique pour détecter, compter et caractériser des cellules de carcinome dans le sang. L'essai peut déceler une cellule épithéliale ou moins dans 1 ml de sang et présente une plus grande sensibilité que le PCR ou l'immunohistochimie classique par 1 à 2 ordres de grandeur. En outre, l'essai facilite la caractérisation biologique et la stadification des cellules de carcinome.
