A highly sensitive assay is disclosed which combines immunomagnetic enrichment with multiparameter flow cytometric and immunocytochemical analysis to detect, enumerate and characterize carcinoma cells in the blood. The assay can detect one epithelial cell or less in 1 ml of blood and has a greater sensitively than conventional PCR or immunohistochemistry by 1-2 orders of magnitude. In addition, the assay facilitates the biological characterization and staging of carcinoma cells.
L'invention porte sur un essai très sensible qui combine un enrichissement immunomagnétique réalisé par cytométrie en flux multiparamétrique et une analyse immunocytochimique. L'essai est utile pour déceler, dénombrer et caractériser les cellules d'un carcinome dans le sang. L'essai peut détecter une cellule épithéliale ou moins dans 1 ml de sang et est associé à une meilleure sensibilité que l'immunohistochimie ou le PCR classique par un facteur de 1 à 2. En outre, l'essai facilite la caractérisation biologique et la stadification des cellules d'un carcinome.
