The present invention relates to methods for increasing the diversity of monoclonal antibodies produced against an antigen. The methods of the invention utilize immunization of a murine host defective in one or more enzymes involved in a post-translational modification of a polypeptide or a modification of a lipid, wherein said modification is exposed on a cell surface. The invention also relates to monoclonal antibodies produced by these methods and which are not produced when a normal mouse is immunized with the same antigen. The invention further relates to compositions comprising these monoclonal antibodies, as well as to such monoclonal antibodies bound or conjugated to a toxin, a detectable marker or to a solid support.
La présente invention concerne un procédé permettant d'accroître la diversité des anticorps monoclonaux produits contre un antigène. Le procédé de l'invention utilisent l'immunisation d'un hôte murin présentant une déficience par rapport à l'une au moins des enzymes impliquées dans la modification post-traductionnelle d'un polypeptide ou la modification d'un lipide, laquelle modification est apparente sur une surface cellulaire. L'invention concerne également des anticorps monoclonaux produits selon ce procédé et qui ne sont pas produits lorsqu'une souris normale est immunisée avec le même antigène. L'invention concerne en outre des compositions comprenant ces anticorps monoclonaux, ainsi que de tels anticorps monoclonaux liés ou conjugués à une toxine, à un marqueur repérable ou à un support solide.
