A method for isolating mesenchymal stem cells from embryo of human or animal is provided. The method comprises following steps: A. a sample derived from a embryo of human or animal is obtained and treated with centrifugation; B. the sample is digested with an enzyme digestion solution to make a single cell suspension, wherein said enzyme digestion solution comprises tripsin-EDTA and collagenase IV; C. mesenchymal stem cell culture medium A is added into the isolated single cells to inoculate and culture the cells; D. the cells are treated with adherence purification and mesenchymal stem cells are obtained by magnetic sorting. A method for isolating the secretion substances of mesenchymal stem cells, and the application of the secretion substances in human cosmetic and healthcare product, and an epidermal cell repairing lotion containing such secretion substances are also provided. During the process of isolating mesenchymal stem cells from embryo of human or animal, most single cells can be obtained by using mixed enzyme digestion solution comprises tripsin-EDTA and collagenase IV as the digestion solution, while reducing the extent of cell damage to the lowest.
Linvention concerne un procédé dextraction de cellules souches interstitielles dembryon humain ou animal comprenant les étapes suivantes : A) une centrifugation est effectuée sur un échantillon à base dembryon humain ou animal; B) un suc digestif enzymatique, composé de pancréatine-EDTA et de collagénase de type IV, est utilisé pour digérer léchantillon et obtenir une suspension de cellules isolées; C) une solution de culture de type A de cellules souches interstitielles est ajoutée aux cellules isolées séparées; les cellules sont inoculées et cultivées; D) les cellules sont purifiées par adhérence et les cellules souches interstitielles sont obtenues via un tri par billes magnétiques. Un procédé dextraction de sécrétion des cellules souches interstitielles, lutilisation sur le plan cosmétique et le plan de la santé de ladite sécrétion, et lutilisation de ladite sécrétion dans la préparation dun liquide réparant les cellules épidermiques sont également proposés. Au cours du processus dextraction de cellules souches interstitielles dembryon humain ou animal, un suc digestif enzymatique composé dun mélange de pancréatine-EDTA et de collagénase de type IV est utilisé. Il permet dobtenir un maximum de cellules isolées et de réduire au maximum les dommages causés aux cellules.
