The present invention provides improved methods for labeling nucleotides. The method of the invention for labeling nucleotides comprises the steps of: reacting a reactive moiety of a linker, which linker is a platinum compound having a stabilizing bridge and two reactive moieties, with an electron donating moiety of a spacer, which spacer comprises a chain having at least four atoms and at least one heteroatom in the chain, which spacer further comprises said electron donating moiety at one end of the chain and a reactive moiety at the other end of the chain; reacting the reactive moiety of said spacer with a label; reacting the other reactive moiety of said linker with a nucleotide. A major advantage of the invention is that all nucleotides can be labeled by the method of the invention, whereas until now the attachment of a label was mostly restricted to one or certain nucleotides.
La présente invention concerne des procédés améliorés de marquage de nuléotides. Le procédé de l'invention destiné au marquage de nucléotides comprend les étapes consistant à faire réagir une fraction réactive d'un lieur, lequel lieur est un composé de platine présentant un pont stabilisateur et deux fractions réactives, avec une fraction donneuse d'électrons d'un espaceur, lequel espaceur comprend une chaîne présentant au moins quatre atomes et au moins un hétéroatome dans la chaîne, ledit espaceur comprenant également ladite fraction donneuse d'électrons au niveau d'une extrémité de la chaîne et une fraction réactive à l'autre extrémité de la chaîne, à faire réagir la fraction réactive dudit espaceur avec une étiquette, à faire réagir l'autre fraction réactive dudit lieur avec un nucléotide. Un avantage majeur de l'invention réside dans le fait que tous les nucléotides peuvent être marqués par le procédé de l'invention, alors que jusqu'à maintenant, l'application d'une étiquette se limitait tout au plus à un nucléotide ou à certains nucléotides.
