The present invention relates to novel methods of making soluble proteins having free cysteines in which a host cell is exposed to a cysteine blocking agent. The soluble proteins produced by the methods can then be modified to increase their effectiveness. Such modifications include attaching a PEG moiety to form pegylated proteins.
L'invention concerne des techniques qui permettent de produire des protéines solubles contenant des cystéines libres. Selon lesdites techniques, on expose une cellule hôte à un agent bloquant la cystéine, puis on modifie éventuellement les protéines solubles obtenues afin d'accroître leur efficacité. Ces modifications peuvent consister à fixer un groupe PEG afin de former des protéines pégylées.
