A method of producing propagation-defective Vesicular Stomatitis Virus (VSV) is provided. The method involves providing a cell that includes an optimized VSV G gene, wherein expression of VSV G protein from the optimized VSV G gene is inducible; and inducing the cell to express VSV G protein from the optimized VSV G gene. The method also involves infecting the induced cell with an attenuated VSV; growing the infected cells in culture; and recovering attenuated VSV from the culture.
La présente invention concerne un procédé de production du virus de la stomatite vésiculaire (VSV) à propagation défectueuse. Le procédé comprend les étapes consistant à mettre à disposition une cellule qui comprend un gène G du VSV optimisé, l'expression de la protéine G du VSV à partir du gène G du VSV optimisé pouvant être induite; et à induire l'expression de la protéine G du VSV dans la cellule à partir du gène G du VSV optimisé. Le procédé comprend également les étapes consistant à infecter la cellule où a eu lieu l'induction avec un VSV atténué; à mettre les cellules infectées en culture; et à récupérer les VSV atténués à partir de la culture.
