Procedures to produce a biologically active human TPO by mammalian cells in in vitro cultures are disclosed. Murine myeloma cells were genetically modified by introducing the gene of human thrombopoietin through a DNA construction which includes an immunoglobulin promoter associated to an immunoglobulin enhancer. The hTPO obtained is useful with methods of stimulating proliferation or development of hematopoietic cells of the megakaryocytic leneage in vitro and in vivo.
L'invention concerne des méthodes permettant de produire une thrombopoïétine (TPO) humaine biologiquement active à partir de cellules mammifères dans des cultures in vitro. Des cellules de myélome murines sont modifiées génétiquement par introduction du gène de la thrombopoïétine humaine dans une construction d'ADN comprenant un promoteur d'immunoglobuline associé à un activateur d'immunoglobuline. La hTPO obtenue est utilisée dans des méthodes de stimulation de la prolifération ou du développement des cellules hématopoïétiques de la lignée mégacaryocytaire in vitro et in vivo.
