The present invention relates to cardiac hypertrophy. More particularly, the present invention defines the molecular events linking calcium stimulation to cardiac hypertrophy. More specifically, the present invention shows that Ca++ stimulation of the hypertrophic response is mediated through MEF2. Thus, the present invention provides methods of treating cardiac hypertrophy as well as transgenic constructs for preparing transgenic animals. Further provided are methods of using transgenic animals of the present invention, or cells isolated therefrom, for the detection of compounds having therapeutic activity toward cardiac hypertrophy.
Cette invention, qui a trait à l'hypertrophie cardiaque, définit, plus précisément, les événements moléculaires liant la stimulation du calcium à l'hypertrophie cardiaque. Elle montre, plus particulièrement, que la stimulation de Ca++ de la réponse hypertrophique est induite par le facteur amplificateur 2 du myocyte (MEF2). Elle porte donc sur des méthodes de traitement de l'hypertrophie cardiaque ainsi que sur des animaux transgéniques ou à des cellules isolées chez ceux-ci aux fins de la détection de composés ayant une action thérapeutique à l'encontre de l'hypertrophie cardiaque.
