The subject invention provides a method for producing an olfactory GPCR in a cell. In general, the methods involve introducing an expression cassette containing a promoter operably linked to a nucleic acid encoding an olfactory PCR into a macroglial cell, e.g., a Schwann or oligodendritic cell, and maintaining the cell under conditions suitable for production of the olfactory GPCR. Also provided is a macroglial cell containing a recombinant nucleic acid encoding an olfactory GPCR, methods of screening for modulators of olfactory GPCR activity, and a kit for producing an olfactory GPCR in a macroglial cell. The invention finds most use in research on flavors and fragrances, and, consequently, has a variety of research and industrial applications.
L'invention concerne un procédé permettant de produire un récepteur couplé à la protéine G (GPCR) olfactif dans une cellule. D'une manière générale, les procédés consistent à introduire une cassette d'expression contenant un promoteur lié de manière fonctionnelle à un acide nucléique codant un GPCR olfactif dans une cellule macrogliale, par exemple une cellule de Schwann ou oligodendritique, et à conserver la cellule dans des états appropriés pour la production du GPCR olfactif. L'invention concerne également une cellule macrogliale contenant un acide nucléique de recombinaison codant un GPCR olfactif, des procédés de criblage de modulateurs de l'activité de GPCR olfactif, ainsi qu'une trousse pour la production d'un GPCR olfactif dans une cellule macrogliale. L'invention concerne plus particulièrement la recherche en matière de saveurs et parfums et, par conséquent divers domaines de la recherche et des applications industrielles.
