The present invention delivers a new concept with respect to the broad definition of alto-antigens that drive graft-versus~tumor (GVT) reactions and/ or graft-versus-host disease (GVHD). The new method for the identification of allo-antigens, which has so far been an unresolved technical problem, furthermore calls for new strategies in immunotherapy. Because the analysis of HLA-binding proteins can now be done with alloantigens for which the corresponding amino~acid sequence is known, the present invention also allows the separation of alto-reactive T cells that only recognize tumor cells from those that mediate GVHD. Antigens defined with the new technology are especially useful for diagnosis and vaccination in cancer and transplantation- related diseases.
La pr~sente invention repose sur un nouveau concept relatif ~ la d~finition large d'allo-antignes entra¹nant des r~actions antitumorales (GVT) et/ou des r~actions de greffe contre hÙte (GVHD).Le nouveau proc~d~ d'identification d'allo-antignes, qui jusqu'~ pr~sent repr~sentait un problme technique non r~solu, ouvre en outre une voie pour de nouvelles strat~gies d'immunoth~rapie. Etant donn~ que l'on peut maintenant effectuer une analyse des prot~ines de liaison HLA avec des allo-antignes pour lesquels la s~quence d'acides amin~s correspondante est connue, l'invention permet ~galement de s~parer les lymphocytes T allor~actifs qui ne reconnaissent que les cellules canc~reuses de ceux qui agissent comme m~diateurs de la r~action greffe contre hÙte. Les antignes d~finis selon le nouveau proc~d~ de l'invention sont particulirement utiles pour le diagnostic et la vaccination dans les maladies canc~reuses et li~es aux transplantations.
