The present invention provides methods of treating, preventing, slowing the progression of, or ameliorating the symptoms of T cell mediated immunological diseases, particularly autoimmune diseases (e.g., autoimmune diabetes (i.e. type 1 diabetes or insulin-dependent diabetes mellitus (IDDM)) and multiple sclerosis) through the use of anti-human CD3 antibodies. The antibodies of the invention of the invention are preferably used in low dose dosing regimens, chronic dosing regimens or regimens that involve redosing after a certain period of time. The methods of the invention provide for administration of antibodies that specifically bind the epsilon subunit within the human CD3 complex. Such antibodies modulate the T cell receptor/alloantigen interaction and, thus, regulate the T cell mediated cytotoxicity associated with autoimmune disorders. Additionally, the methods of the invention provide for use of anti-human CD3 antibodies modified such that they exhibit reduced or eliminated effector function and T cell activation as compared to non-modified anti-human CD3 antibodies.
La présente invention concerne des procédés permettant de traiter, prévenir, ralentir la progression ou améliorer les symptômes de maladies immunologiques médiées par les lymphocytes T, en particulier des maladies auto-immunes (par exemple le diabète auto-immun (c'est-à-dire le diabète de type 1 ou le diabète sucré insulinodépendant (DSID)) et la sclérose en plaques), en utilisant des anticorps anti-CD3 humain. Les anticorps de l'invention sont utilisés de préférence dans des schémas posologiques à faible dose, des schémas posologiques chroniques ou des schémas impliquant un rappel après une certaine durée. Il est prévu selon les procédés de l'invention l'administration d'anticorps qui se lient spécifiquement à la sous-unité epsilon du complexe CD3 humain. Ces anticorps modulent l'interaction entre le récepteur des lymphocytes T et l'alloantigène et, de ce fait, ils régulent la cytotoxicité médiée par les lymphocytes T qui est associée aux pathologies auto-immunes. En outre, il est prévu selon les procédés de l'invention d'utiliser des anticorps anti-CD3 humain modifiés de façon à présenter une fonction effectrice et une activation des lymphocytes T réduite ou supprimée, comparés à des anticorps anti-CD3 humain non modifiés.
