The present invention relates to the use of irinotecan or a derivative thereof for the preparation of a pharmaceutical composition for treating cancer, especially, colorectal cancer, cervical cancer, gastric cancer, lung cancer, malignant glioma, ovarian cancer, and pancreatic cancer in a patient having a genotype with a variant allele which comprises a polynucleotide in accordance with the present invention. Preferably, a nucleotide deletion, addition and/or substitution comprised by said polynucleotide results in an altered expression of variant allele compared to the corresponding wild type allele or an altered activity of the polypeptide encoded by the variant allele compared to the polypeptide encoded by the corresponding wild type allele. Finally, the present invention relates to a method for selecting a suitable therapy for a subject suffering from cancer, especially, colorectal cancer, cervical cancer, gastric cancer, lung cancer, malignant glioma, ovarian cancer, and pancreatic cancer.
L'invention concerne l'utilisation d'irinotécan ou d'un dérivé de celui-ci dans la préparation d'une composition pharmaceutique destinée à traiter le cancer, notamment les cancers colorectal, cervical et gastrique, le cancer du poumon, le gliome malin, le cancer de l'ovaire et le cancer du pancréas chez un patient présentant un génotype avec un allèle variant qui comprend un polynucléotide selon cette invention. De préférence, la suppression, l'ajout et/ou la substitution du nucléotide compris dans ce polynucléotide mène à l'expression d'un allèle variant comparé à un allèle correspondant de type sauvage ou à une activité modifiée du polypeptide codé par l'allèle variant en comparaison au polypeptide codé par l'allèle de type sauvage correspondant. L'invention concerne finalement un procédé pour sélectionner une thérapie appropriée destinée à un sujet souffrant du cancer, notamment des cancers colorectal, cervical ou gastrique, le cancer du poumon, le gliome malin, le cancer de l'ovaire et le cancer du pancréas.
