The present invention relates to the finding that receptors for the neurotransmitter 5-HT of the 5-HT2B class are located on the human colon, and that their activation potentiates neuronally-mediated responses, and are thus causative in the abnormal motility and pain associated with IBS. The invention provides a method of treatment of IBS which comprises providing to a patient in need of treatment an effective amount of a 5HT2B receptor antagonist which acts on 5HT2B receptors located in the colon of said patient. Such antagonists may be administered in the form of compositions adapted to be delivered to the colon, such as depot formulations and suppositories. Assay methods for the development of compounds for the treatment of IBS are also provided.

L'invention concerne la découverte selon laquelle des récepteurs destinés au neurotransmetteur 5-HT de la classe 5-HT¿2B? sont situés sur le côlon humain et selon laquelle l'activation de ceux-ci potentialise des réponses induites par voie neuronale, ces récepteurs étant, par conséquent, responsables de motilité et de douleurs anormales associées au syndrome IBS. L'invention concerne également une méthode de traitement du syndrome IBS consistant à administrer à un patient nécessitant un traitement une quantité efficace d'un antagoniste du récepteur 5HT2B agissant sur des récepteurs 5HT2B situés dans le côlon du patient. De tels antagonistes peuvent être administrés sous forme de compositions conçues pour être administrées dans le côlon, telles que des formulations dépôt et des suppositoires. L'invention concerne enfin des méthodes d'analyse destinées à l'élaboration de composés conçus pour le traitement du syndrome IBS.
