The invention relates to the use of parathyroid hormone-related protein (PTHrP) in the prevention of hypertrophy in chondrogenic cells for cartilage replacement. A method for engineering three dimensional cartilage constructs from chondrogenic cells is provided, said method comprising a step of treating the chondrogenic cells or immature constructs with PTHrP to regulate hypertrophy. Also provided are: three dimensional cartilage produced by the method of the invention, and an engineered cartilage construct comprising chondrogenic cells and a bioactive scaffold capable of controlled release of PTHrP. In addition, the invention provides a method for the treatment of osteoarthritis.
L'invention concerne l'utilisation de protéine relative à l'hormone parathyroïde (PTHrP) dans la prévention de l'hypertrophie dans des cellules chondrogéniques, aux fins de remplacement du cartilage. L'invention concerne également un procédé de ingénierie destinés à des constructions de cartilage tridimensionnelles à partir de cellules chondrogéniques, ce procédé comprenant un étape consistant à traiter les cellules chondrogéniques ou des constructions immatures au moyen de PTHrP, de manière à réguler l'hypertrophie. L'invention concerne, en outre: un cartilage tridimensionnel produit au moyen du procédé selon l'invention et une construction de cartilage fabriquée comprenant des cellules chondrogéniques et un échafaudage bioactif capable de libérer de manière commandée PTHrP. De plus, l'invention concerne une méthode de traitement de l'ostéoarthrite.
