The present invention provides methods, compositions and kits for using an RNA polymerase for making transcription products corresponding to a target sequence by obtaining circular single-stranded DNA transcription substrates using a promoter primer that encodes one strand of a double-stranded promoter. The invention has broad applicability for research, diagnostic and therapeutic applications, such as preparing cDNA corresponding to mRNA, making sense or anti-sense probes, detecting gene- or organism-specific sequences, or making RNAi.

L'invention concerne des procédés, des compositions et des trousses permettant d'utiliser une ARN polymérase afin d'obtenir des produits de transcription correspondant à une séquence cible par obtention de substrats de transcription d'ADN monocaténaire circulaire au moyen d'une amorce de promoteur qui code un brin d'un promoteur bicaténaire. L'invention présente de nombreuses possibilités d'application dans la recherche, le diagnostic et la thérapie, telles que la préparation d'ADNc correspondant à ARNm, la production de sondes sens et anti-sens, la détection de séquences spécifiques à un gène ou à un organisme ou la production d'ARNi.
