Disclosed are methods of 6-O-sulfating glucosaminyl N-acetylglucosamine residues (G1cNAc) in a polysaccharide preparation and methods of converting anticoagulant-inactive heparan sulfate to anticoagulant-active heparan sulfate and substantially pure polysaccharide preparations may by such methods. Also disclosed is a mutant CHO cell which hyper-produces anticoagulant-active heparan sulfate. Methods for elucidating the sequence of activity of enzymes in a biosynthetic pathway are provided.
L'invention porte sur des procédés de sulfatation en 6-O de groupes de glucosaminyl N-acétylglucosamine (G1cNAc) dans une préparation de polysaccharides, et sur des procédés de conversion de sulfate d'héparan inactif comme anticoagulant en sulfate d'héparan actif comme anticoagulant, et sur des préparations sensiblement pures de polysaccharides obtenues à l'aide desdits procédés. L'invention porte également sur une cellule mutante de CHO hyperproductrice d'héparan actif comme anticoagulant, et sur un procédé d'élucidation de la séquence d'activité d'enzymes dans un mécanisme de biosynthèse.
