The present invention relates to the field of stem cell culture, in particular undifferentiated stem cell culture and to methods for derivation and propagation of such cells. More particularly, the invention relates to derivation and propagation of undifferentiated HES cells on human feeder layers and/or in the absence of a feeder layer.The human feeder layers may be selected from the group including human fetal muscle (HFM), human fetal skin (HFS), human adult fallopian tubal (HAFT) fibroblasts and human adult skin cells. They may be cultured in the presence of a suitable medium selected from the group including Human Embyonic Stem Cell (HES), Knockout (KO), or Human Feeder (HF) medium supplemented with or without human serum.

L'invention se rapporte au domaine de la culture de cellules souches, en particulier à la culture de cellules souches non différenciées, ainsi qu'à des procédés de dérivation et de propagation desdites cellules. Plus particulièrement, l'invention se rapporte à la dérivation et à la propagation de cellules souches embryonnaires humaines (HES) sur des couches nourricières humaines et/ou en l'absence d'une couche nourricière. Les couches nourricières humaines peuvent être sélectionnées dans le groupe comprenant un muscle foetal humain (HFM), de la peau foetale humaine (HFS), des fibroblastes des trompes utérines d'une femme adulte (HAFT) et des cellules de la peau d'une personne adulte. Lesdites couches peuvent être cultivées en présence d'un milieu approprié sélectionné dans le groupe comprenant des cellules souches embryonnaires humaines (HES), un milieu d'inactivation (KO) ou un milieu nourricier humain (HF) avec ou sans sérum humain.
