A method of enhancing the isolation of ribonucleic acids from samples of biological or cellular material is disclosed which uses a solution of an Agent during binding of nucleic acids onto a solid phase binding material which, after elution, enhances the recovery of RNA. Washing the solid phase and eluting the nucleic acid produces RNA in enhanced yield and/or purity. The use of the new method allows RNA to be captured and released in a form suitable for downstream processing in under five minutes. Preferred Agents according to the invention comprise monomeric, oligomeric, dendrimeric and polymeric organic compounds having multiple positive charges.
L'invention concerne un procédé permettant de faciliter l'isolation d'acides ribonucléiques dans des échantillons de matériau biologique ou cellulaire. Ledit procédé consiste à utiliser une solution d'un agent pendant la liaison d'acides nucléiques à un matériau de liaison en phase solide qui, après élution, favorise la récupération d'ARN. Le nettoyage de la phase solide et l'élution de l'acide nucléique permettent de produire un ARN ayant un rendement et/ou une pureté améliorés. Ce nouveau procédé permet de capturer et de libérer l'ARN sous une forme convenant au traitement en aval en mois de cinq minutes. Les agents préférés selon l'invention renferment des composés organiques monomères, oligomères, dendrimères et polymères possédant des charges positives multiples.
