The methods described herein provide for ways of modulating, specifically inhibiting, integrin activation. The methods include identifying compounds such as naturally occurring and non-naturally occurring polypeptides and small molecules which bind to the .beta.A domain of an integrin thus mimicking the contact betwen the CD loop with .beta.A domain. Contacting the .beta.A domain in this manner locks the integrin such that it is unable to be activated.

L'invention concerne des méthodes permettant d'engendrer des voies de modulation, notamment, d'inhibition de l'activation de l'intégrine. Ces méthodes consistent à identifier des composés, tels que des polypeptides qui se produisent naturellement ou non et des petites molécules qui se lient au domaine .beta.A d'une intégrine, ce qui permet d'imiter le contact entre la boucle CD et le domaine .beta.A. La mise en contact du domaine .beta.A de cette manière permet de bloquer l'intégrine, afin qu'elle ne puisse pas être activée.
