A method of marking a material and subsequently detecting that it has been marked is disclosed, comprising adding or applying a marker comprising a nucleic acid tag to the material, sampling a portion of the material containing such marker, and detecting the presence of nucleic acid tag in the sample. The method is characterised in that the quantity of the nucleic acid tag present in the sample is determined to provide an indication of the quantity of marker present in the material. The nucleic acid tag may comprise one or more identifying sequences, and the tag is amplified by means of a nucleic acid amplification reaction. Also disclosed are a method of marking a material with a nucleic acid tag, a method of detecting whether a material has been marked by a marker comprising a nucleic acid tag, a method of detecting the quantity of a marker comprising a nucleic acid tag in a material that has previously been marked, a method of marking a material and subsequently determining the quantity of marker in the material, and a method of and a kit for determining which particular nucleic acid tag from a known pool of nucleic acid tags has been used to mark a material.
L'invention concerne un procédé de marquage d'un matériau et ultérieurement de détection dudit marquage sur le matériau. Le procédé consiste à ajouter ou à appliquer au matériau un marqueur comprenant une étiquette formée d'un acide nucléique, à prélever un échantillon dudit matériau contenant un tel marqueur , puis à détecter la présence de l'étiquette d'acide nucléique dans ledit échantillon. Le procédé est caractérisé par le fait que la quantité de l'étiquette d'acide nucléique présente dans ledit échantillon est déterminée de manière à fournir une indication sur la quantité de marqueur présent dans le matériau. Ledit fragment peut comporter une ou plusieurs séquences d'identification, et il est amplifié au moyen d'une quantification par RT-PCR. L'invention concerne également un procédé de marquage d'un matériau à l'aide d'une étiquette d'acide nucléique; un procédé permettant de savoir si un matériau a été marqué par un marqueur qui contient une étiquette d'acide nucléique; un procédé permettant de détecter la quantité d'un marqueur comprenant une étiquette d'acide nucléique dans un matériau préalablement marqué; un procédé de marquage d'un matériau, ledit procédé permettant, par la suite, de détecter la quantité de marqueur dans ledit matériau; et un procédé et une trousse permettant de déterminer quelle étiquette d'acide nucléique spécifique, issue d'un groupe d'étiquettes d'acide nucléique connu, a été utilisée pour le marquage du matériau.
