Provided are methods and processes for producing glycoproteins by eukaryotic cell culture which provide a glycoprotein product containing oligosaccharides terminating in one or more sialic residues by inhibiting sialidase expression and/or activity in the cell culture. Sialidase expression and/or activity was shown to be inhibited by the addition of a trace element, e.g. zinc or cobalt, or insulin-like growth factor 1 (IGF-1) to the cell culture medium.

Linvention concerne des procédés et des processus de production de glycoprotéines par culture de cellules eucaryotes, qui fournissent un produit de glycoprotéine contenant des oligosaccharides se terminant par un ou plusieurs résidus sialiques par inhibition de lexpression et/ou de lactivité de la sialidase dans la culture de cellules. Il a été montré que lexpression et/ou lactivité de la sialidase sont inhibées par laddition dun élément trace, p. ex. le zinc ou le cobalt, ou du facteur de croissance analogue à linsuline 1 (IGF-1), au milieu de culture de cellules.
