Novel methods of testing the immunogenicity of variant antigens are provided. In particular, methods based on the use of transgenic animals are provided, wherein the transgenic animal is tolerised to a particular antigen and is then exposed to variants of the antigen and immune responses are determined. In one embodiment the transgenic animal is a mouse which is transgenic for human MHC class II molecules and the immunogenicity of libraries of variant antibodies are tested.

La présente invention concerne de nouveaux procédés pour tester l'immunogénicité d'antigènes de variantes. Elle concerne notamment des procédés basés sur l'utilisation d'animaux transgéniques, selon lesquels l'animal transgénique est rendu tolérant à un antigène particulier, puis est ensuite exposé à des variantes de l'antigène et des réponses immunitaires sont déterminées. Dans un mode de réalisation, l'animal transgénique est une souris qui est transgénique pour des molécules MHC de classe II humaines et l'immunogénicité de banques d'anticorps de variantes est testée.
