Methods of therapy are provided for treating a subject for a solid tumor that comprises carcinoma cells expressing the CD40 cell-surface antigen. The methods comprise administering a therapeutically effective amount of an antagonist anti-CD40 antibody or antigen-binding fragment thereof to a subject in need thereof. The antagonist anti-CD40 antibody or antigen-binding fragment thereof is free of significant agonist activity, but exhibits antagonist activity when the antibody binds a CD40 antigen on a human CD40-expressing cell. Binding of the anti-CD40 antibody or suitable antigen-binding fragment thereof beneficially inhibits growth of solid tumors via ADCC-dependent killing of the CD40-expressing carcinoma cells.
L'invention porte sur des procédés thérapeutiques permettant de traiter un sujet atteint d'une tumeur solide qui contient des cellules de carcinome exprimant l'antigène de surface cellulaire CD40. Ces procédés consistent à administrer une quantité thérapeutiquement efficace d'un anticorps anti-CD40 antagonsite ou un fragemnt à liaison antigène à un sujet qui en a besoin. Cet anticorps anti-CD40 antagonsite ou fragment à liaison antigène ne présente pas d'activité agoniste significative, mais présente une activité antagonsite lorsque l'anticorps se lie à un antigène CD40 sur une cellule d'expression CD40 humaine. La liaison de l'anticorps anti-CD40 ou du fragment à liaison antigène convenable inhibe de manière bénéfique la croissance des tumeurs solides par suppression fondée sur ADCC des cellules de carcinome exprimant CD40.
