Provided are methods of using gelsolin and active fragments thereof to neutralize, treat or prevent the pathogenic effects of lipopolysaccharide (LPS) endotoxins released from gram-negative bacteria, including massive activation of inflammatory response in a patient and the resulting lethal septic shock. The provided gelsolin binds LPS from various bacteria with high affinity, decreasing circulating LPS and neutralizes its deleterious biological effects. Consequently, the provided gelsolin replacement therapy offers a method for the prevention of LPS-induced mortality in the patient.
La présente invention concerne des procédés permettant d'utiliser la gelsoline et certains des ses fragments actifs pour neutraliser, traiter pou prévenir les effets pathogènes d'endotoxines à lipopolysaccharides (LPS) libérées par les bactéries Gram négatives, y compris l'activation massive de réponse inflammatoire chez un patient, et le choc septique létal en résultant. La gelsoline de l'invention se lie aux LPS de diverses bactéries avec une grande affinité, ce qui diminue les LPS en circulation et en neutralise les effets biologiques délétères. Il en résulte que cette thérapie de substitution par la gelsoline constitue un mode de prévention de la mortalité induite par les LPS chez le patient.
