The present invention provides methods of modulating mammalian, particularly human, stem cell and progenitor cell differentiation to regulate and control the differentiation and maturation of these cells along specific cell and tissue lineages. The methods of the invention relate to the use of certain small organic molecules to modulate the differentiation of stem cell populations along specific cell and tissue lineages, particularly embryonic- like stem cells originating from a postpartum placenta or stem cells isolated form sources such as cord blood. The invention also relates to the treatment or prevention of myelodysplastic syndrome or myeloproliferative syndrome, or symptoms thereof, comprising administration of JNK or MKK inhibitors, alone or in combination, as well as with or without the use of unconditioned cells or cells conditioned in accordance with other aspects of the invention. Finally, the invention relates to the use of such differentiated stem cells in transplantation and other medical treatments.
L'invention concerne des procédés destinés à moduler la différenciation de cellules souches et de cellules progénitrices mammaliennes, et notamment humaines, en vue de réguler et de surveiller la différenciation et la maturation de ces cellules le long de lignées cellulaires et tissulaires spécifiques. Les procédés de l'invention concernent l'utilisation de certaines molécules organiques de petite dimension en vue de moduler la différenciation de populations de cellules souches le long de lignées tissulaires spécifiques, notamment des cellules souches de type embryonnaires provenant du placenta du post-partum ou des cellules souches isolées de sources telles que du sang de cordon ombilical. L'invention concerne également le traitement ou la prévention du syndrome myélodysplasique ou du syndrome myéloprolifératif, ou de symptômes de ceux-ci, qui consiste à administrer des inhibiteurs de JNK ou MKK, seuls ou en combinaison, avec ou sans utilisation de cellules non traitées ou de cellules traitées conformément à d'autres aspects de l'invention. Enfin, l'invention concerne l'utilisation de ces cellules souches différenciées dans la transplantation et d'autres traitements médicaux.
