This invention provides methods for modulating gene expression at the transcriptional level. In particular, the methods involve tethering a transcriptional coactivator to a DNA binding domain that is specific for a target nucleic acid sequence and contacting the coactivator with a transcription factor. The transcription factor triggers or represses transcription mediated by the coactivator. Methods for identifying compounds that are able to modulate gene expression are also provided.

L'invention concerne des procédés servant à moduler l'expression génétique au niveau de la transcription. Ces procédés consistent, en particulier, à relier un coactivateur de transcription à un domaine de fixation d'A D N spécifique pour une séquence cible d'acides nucléiques et à mettre en contact ce coactivateur avec un facteur de transcription. Ce dernier déclenche ou réprime la transcription provoquée par le coactivateur. Elle concerne également des procédés servant à identifier des composés capables de moduler l'expression génétique.
