The present invention relates to compounds which inhibit or antagonize the binding of methylglyoxal (MG) and/or other reactive carbonyl species (RCS) to an arginine- or lysine- containing protein, preferably an arginine- or lysine-containing cellular protein, such as a sodium ion channel, e.g. the sodium ion channel Na(v)1.8. Preferred scavenger compounds are peptides comprising several or multiple repeats of the amino acid sequence motif GIy- Glu-X-Pro (GEXP), wherein X is Arg or Lys, and pharmaceutical compositions thereof. The present invention furthermore relates to the use of the compounds as scavenger or antagonists of methylglyoxal and/or related reactive carbonyl species (RCS). The present invention furthermore relates to the use of the compounds for the prevention and/or treatment of pain, hyperalgesia and pain related diseases, in particular pain and/or hyperalgesia caused by or associated with methylglyoxal and/or reactive carbonyl species (RCS).
La présente invention concerne des composés qui inhibent ou jouent le rôle d'antagonistes dans la liaison du méthylglyoxal (MG) et/ou d'autres espèces de carbonyle réactives (RCS) à une protéine contenant de l'arginine ou de la lysine, de préférence une protéine cellulaire contenant de l'arginine ou de la lysine, telle qu'un canal sodique, par exemple, le canal sodique Na(v)1.8. Les composés de piégeage préférés sont des peptides comprenant plusieurs ou de multiples répétitions du motif de séquence d'acides aminés Gly-Glu-X-Pro (GEXP), où X est Arg ou Lys. L'invention se rapporte également à des compositions pharmaceutiques de ces composés. La présente invention concerne en outre l'utilisation desdits composés en tant qu'agents de piégeage ou antagonistes du méthylglyoxal et/ou d'espèces de carbonyle réactives (RCS) apparentées. La présente invention a également trait à l'utilisation des composés pour la prévention et/ou le traitement de la douleur, de l'hyperalgésie et de maladies auxquelles s'associe la douleur, en particulier la douleur et/ou l'hyperalgésie causée par ou associée au méthylglyoxal et/ou à des espèces de carbonyle réactives (RCS).
