Mismatch Repair Detection (MRD), a novel method for DNA-variation detection, utilizes bacteria to detect mismatches by a change in expression of a marker gene. DNA fragments to be screened for variation are cloned into two MRD plasmids, and bacteria are transformed with heteroduplexes of these constructs. Resulting colonies express the marker gene in the absence of a mismatch, and lack expression in the presence of a mismatch. MRD is capable of detecting a single mismatch within 10 kb of DNA. In addition, MRD can analyze many fragments simultaneously, offering a powerful method for high-throughput genotyping and mutation detection.

L'invention concerne une détection de réparation de mésappariement (MRD) qui est un nouveau procédé de détection de variation d'ADN mettant en oeuvre des bactéries, aux fins de détection de mésappariements par un changement dans l'expression d'un gène marqueur. Des fragments d'ADN à balayer aux fins de variation sont clonés dans deux plasmides de détection MRD et des bactéries sont transformées au moyen d'hétéroduplexes de ces constructions. Des colonies obtenues expriment le gène marqueur en l'absence d'un mésappariement et n'ont pas d'expression en présence d'un mésappariement. La détection MRD est capable de détecter un mésappariement unique dans 10 kb d'ADN.
