A method of diagnosis or prognosis of a disease or characterisation of a genotype or genetic sequence which method comprises providing a nucleic acid sample and determining the nature of one or more genetic variants of an MMP-9 gene. Also described are oligonucleotides which hybridise to variants of a polymorphic sequence in the MMP-9 gene or its reverse complement. The method and oligonucleotides are of particular use for the diagnosis and prognosis of diseases characterised by metalloproteinase mediated remodelling, such as atherosclerosis, tumour invasion and metastasis, or inflammatory disease.

L'invention se rapporte à une méthode visant à établir un diagnostic ou un pronostic de maladie ou une caractérisation d'un génotype ou d'une séquence génétique. Ladite méthode consiste à utiliser un échantillon d'acide nucléique et à définir la nature d'un ou de plusieurs variants d'un gène de MMP-9. L'invention se rapporte également à des oligonucléotides qui s'hybrident à des variants d'une séquence polymorphe dans le gène de MMP-9 ou son complément inverse. Ladite méthode et lesdits oligonucléotides s'avèrent particulièrement utiles pour établir le diagnostic ou le pronostic de maladies caractérisées par une transformation vasculaire à médiation assurée par des métalloprotéases, du type athérosclérose, métastase et invasion tumorale ou maladie inflammatoire.
