The present invention discloses a double transgenic fly that expresses both human Tau protein and the human A.beta.42 peptide of human amyloid-.beta. precursor protein (APP). The double transgenic flies of the present invention display a synergistic altered phenotype as compared to the altered phenotype displayed by transgenic flies expressing either human Tau or human A.beta.42 alone. Thus, the flies provide for models of neurodegenerative disorders, such as Alzheimer's disease. The invention further discloses methods for identifying therapeutic compounds to treat neurodegenerative disorders using the double transgenic flies.
L'invention concerne une mouche doublement transgénique exprimant à la fois une protéine humaine Tau et le peptide A.beta.42 humain de la protéine précurseur de l'amyloïde-.beta. (APP) humaine. Les mouches doublement transgéniques selon l'invention présentent un phénotype modifié synergique comparé au phénotype modifié que présentent les mouches transgéniques exprimant seulement soit la Tau humaine, soit le A.beta.42 humain. Ces mouches constituent des modèles de troubles neurodégénératifs, tels que la maladie d'Alzheimer. L'invention concerne également des procédés permettant d'identifier des composés thérapeutiques destinés au traitement des troubles neurodégénératifs, au moyen desdites mouches doublement transgéniques.
