Bacteria consume a variety of biomass-derived substrates and produce ethanol. Hydrogenase genes have been inactivated m Thermoanaerobacterium saccharolyticum to generate mutant strains with reduced hydrogenase activities. One such mutant strain with both the ldh and hydtrA genes inactivated shows a significant increase in ethanol production. Manipulation of hydrogenase activities provides a new approach for enhancing substrate utilization and ethanol production by biomass-fermenting microorganisms.

L'invention concerne des bactéries consommant une variété de substrats dérivés de biomasse et produisant de l'éthanol. Des gènes d'hydrogénase ont été inactivés dans Thermoanaerobacterium saccharolyticum pour générer des souches mutantes ayant des activités d'hydrogénase réduites. Une telle souche mutante comportant les deux gènes ldh et hydtrA inactivés, affiche une augmentation significative de production d'éthanol. Une manipulation des activités d'hydrogénase fournit une nouvelle approche pour amplifier l'utilisation de substrat et la production d'éthanol par des micro-organismes fermentant la biomasse.
