The present invention relates to modified ADAMTS4 proteins having improved stability comparing to the corresponding native, unmodified proteins. The modified ADAMTS4 proteins can be expressed and isolated in large quantities, thus allowing further characterization of the proteins, such as crystallographic and enzyme kinetic studies. The purified, stable proteins would also facilitate the production of antiADAMTS antibodies and the development of inhibitors to ADAMTS enzymes.
La présente invention concerne des protéines ADAMTS4 modifiées présentant une plus grande stabilité par rapport aux protéines non modifiées natives correspondantes. Ces protéines ADAMTS4 modifiées peuvent être exprimées et isolées en grandes quantités, ce qui permet de poursuivre la caractérisation des protéines consistant notamment à réaliser des études cinétiques enzymatiques et cristallographiques. Les protéines stables purifiées permettent également de faciliter la production d'anticorps anti-ADAMTS et le développement d'inhibiteurs d'enzymes ADAMTS.
