2070682 9207003 PCTABS00012 The invention relates to a method for preventing the interaction of growth factors or cytokines with their specific cell receptors in mammals. The method comprises administering to the mammal a therapeutic amount of a chemically modified .alpha.2-macroglobulin having the ability to form a .alpha.2-macroglobulin-growth factor or -cytokine complex with a given growth factor or cytokine. The complex preferentially undergoes recepto-medicated modification, preferably endocytosis by .alpha.2-macroglobulin receptors, thereby reducing the availability of the active growth factor or cytokine to its specific cell surface receptor. The complexes are particularly useful in the treatment of endotoxic or septic shock through the scavenging of TNF-.alpha..
